Age: Adult and young Miis cannot fall in love. A child is any Mii under the age of 18, and you can adjust the ages in the Edit Mii menu.
Real Family: Miis that are set as close family members cannot form romantic relationships with each other. However, this limitation can be overridden by changing their relationship status to “partner” in the Edit Mii menu.
Friendship: Miis must become friends before they can develop romantic feelings.

Use the Edit Mii menu to confirm that their dating preferences match, that they're from the same age group, and that they're not real-world family members. To access that menu, select a Mii by pressing A, then press Y to bring up the Mii menu, then select Edit Mii at the bottom.
Encourage frequent interaction between Miis. If the Miis have been separated for a while, hold A to lift one and place them next to the other to start a conversation. This will eventually cause the Miis to become friends, a requirement for romance and continued interaction will lead to love.
Be patient. It can take time for Miis to develop romantic feelings for each other, even when they are compatible. But if you're diligent about getting your Miis to interact, you should eventually get to events that lead to love.
