Of course, Monkey D. Luffy is the primary user of Gear 5 in One Piece. Luffy was the first person seen to use this power, and he did so in the Wano Country Arc. After his massive battle against Kaido, Luffy faced certain death and even his voice disappeared completely, signifying his end. However, it was at this point Luffy's mind and body caught up with his abilities, and thus the Nika Fruit finally awoke.

Jewelry Bonney is a very important character in the Final Saga of One Piece. In the Egghead arc, she took the front seat by becoming the arc's deuteragonist, and this is where she harnessed the power of Gear 5 for the very first time. Bonney was able to use Gear 5, thanks to his own Devil Fruit, Toshi Toshi no Mi.

Before Luffy, there was another user of Gear 5 in One Piece, and that is Joy Boy. Joy Boy existed a long time ago during the Void Centurywhich is confirmed in the story's Egghead arc. He was the former user of Hito Hito no Mi, Model: Nikaand he used its powers exceedingly well. How Joy Boy ate the Devil Fruit is still unknown, but it is already clear in the story that he actually harnessed the abilities of this fruit and that he was the former Nika.
With his abilities he took on the twenty kings of the world, and this battle raged for many years. Eventually Joy Boy lost the battle and died, however his will has now been inherited by none other than Luffy. Fans will definitely get a proper look at Joy Boy with the Gear 5 when his own flashback begins, which should be sometime in the final saga.

Long before Joy Boy, the original sun god Nika existed in the One Piece world as well. This was confirmed in the Elbaf arc of One Piece. Years ago, fans thought that Joy Boy was the first sun god Nika, but it is now clear the first Nika appeared during the First World, which was about 3,000 years ago.
It was there that the sun god Nika was first fought, which means that particular deity was the first user of Gear 5 in One Piece. Very little is known about this individual, but according to Harley, Nika went up against the Hell Serpent and the Earth God together, and their battle devastated the entire world. It was many years after the destruction of the entire world that life took shape again, and that's where the other world, where Joy Boy arose, took place.
