Resident Evil Requiem has a mysterious challenge called “The Final Puzzle” that's worth 20,000 completion points, but no one has been able to solve it yet, and it's not for lack of trying. In fact, the community has pooled its knowledge into a dedicated subreddit that has one goal: to get to the bottom of this bizarre rabbit hole.

The first step becomes clear when you escape the basement of Rhodes Hill Chronic Care Center. Grace narrowly escapes the girl's clutches when the elevator crashes down, severing her hand, which you can pick up after the cutscene ends. It's not immediately clear what the hand is used for, but when you leave the water treatment clinic you'll notice that it's disposable, so it must be used for the first half of the game.
As broken down in an extensive thread by u/Naeloah on r/TheFinalPuzzle, it was found that the hand can be analyzed with a laser microscope to reveal a hidden message: “Let's play,” followed by the sequence 'GGC AAG AUA ACG UGU CAU.' It was then thought that each letter could correspond to the symbols of the sun, moon and star used to open the various units around the quarter block clinic.
Synth Potato, who lights up as an aeronautical engineer, noticed that the hourglass in Victor's office had another hidden message, “U = 380,000” and that “G = 150,000,000” was written on the side of the blood analyzer. The final clue was revealed in a file on Grace's blood test, “C = 4.2 ly.”
As it turns out, “ly” stands for light years. 4.2 light years from Earth is a star, 380,000 km away is the moon and 150,000,000 km away is the sun. It means:

U = The Moon.
That leaves “A”, which was actually resolved much later in the game during Leon Kennedy's return to Raccoon City. After defeating Mr. X on the way to the orphanage, fans found the head of a Toy Uncle bobblehead in the middle of the basketball court, which matched the body in the Kendo's Shop. Interestingly, if you kick the basketball into the fallen hoop, you can interact with the body to get another clue: “Just ignore A.”
So, the sequence is now “GGC GUC GUG UCU”, which, when fed into the lead scientist's office cabinet, plays the sound of a child laughing. The only clue that The Final Puzzle gives you in the challenge menu is that you need to “Let the cute couple hear the voice”, so the working theory right now is that you need to bring two characters (or enemies) into this room and make them listen to the child's laughter.
Other translations may provide insight into who this couple is; as noted by u/Map_Goblin, it ranges from “adorable duo” to “little friends”, “adorable couple”, “loving couple”, “couple of angels”, “two of us” (both female), “cute couple” and “lovely two people”. This seems to rule out the idea that Leon and Grace must complete the puzzle (in fact, Leon doesn't seem to be able to interact with it at all), but who exactly the “cute couple” is is unclear.
Some have tried bringing Emily with Grace to get the laughs going, but it didn't work, and it seems impossible to get both singing zombies into the room. Another idea that was floated was to see the “kids” in the model viewer after hearing the laughter, but that didn't work either. But the laugh might just be part of the code, as the puzzle box can still be interacted with after hearing it. You can read a full list of everything the community has tried here.
A prevailing theory right now is that the laughter is connected to the orphanage later in the game, and that it may play a key role in solving The Final Puzzle; one idea is that when the children beat the beds while looking for the little girl, they do it in a pattern. Some believe this could be Morse code, tying in to the “Let's play” clue, but it's unclear what meaning it might have at this point, if any. Then there is the carpet, which has star, sun and moon symbols surrounded by letters.
Interestingly, when Leon falls into the sewers of the destroyed orphanage, there is an empty room with panels colored red, green and blue for “no apparent reason”, which has raised all sorts of ideas that it could be another step in completing the challenge. But again, no one is sure how it connects, if it even does.
If nothing else, Capcom is really making us work for those 20,000 completion points. But what lies on the other side of all these cryptic puzzles no one knows – let's just hope it's worth all the trouble.
